Biochemical Integration → Fatty Acid Synthesis
Fatty Acid Synthesis
Storing carbon and reducing power during abundance.
01The idea
Fatty acid synthesis is not the reverse of β-oxidation. It solves a different physiological problem. When nutrients are abundant, ATP demand is met, reducing power is high, and carbon must be stored safely.
This is about capacity management, not immediate fuel use.
THREAD · Regulation vs. capacity
02Build the model
AOxidation vs. synthesis: deliberate separation
| Feature | β-Oxidation | Fatty acid synthesis |
|---|---|---|
| Location | Mitochondria | Cytosol |
| Redox | Produces NADH/FADH₂ | Consumes NADPH |
| State | Fasting / stress | Fed / insulin-high |
| Purpose | Energy extraction | Energy storage |
Cells enforce directionality by architecture, not hope.
BCarbon source: from mitochondria to cytosol
Acetyl-CoA is produced in mitochondria, but synthesis occurs in the cytosol, and acetyl-CoA cannot cross the mitochondrial membrane directly. The solution: acetyl-CoA becomes citrate, citrate is exported, and acetyl-CoA is regenerated in the cytosol.
CNADPH: the reducing currency of synthesis
Fatty acid synthesis requires NADPH, not NADH. Major sources: the pentose phosphate pathway, and malic enzyme (context-dependent).
This is why PPP belongs before this lesson.
DThe fatty acid synthase complex: efficiency through integration
Fatty acid synthesis occurs via a large, multi-enzyme complex, in repeated reaction cycles, with two-carbon additions per cycle. You do not need step memorization.
EDirectionality: why this is not reversal
Despite superficial similarity, synthesis and β-oxidation use different enzymes, different cofactors, different locations, and different regulation.
This is a universal metabolic principle.
FHormonal control: state determines direction
| Activated by | Suppressed by |
|---|---|
| High insulin | Glucagon |
| High glucose | Epinephrine |
| High citrate | Low energy state |
Substrate availability alone is insufficient.
GTissue context: where fat is made
Fatty acid synthesis occurs primarily in the liver and adipose tissue. Muscle uses fat but does not significantly synthesize it.
HIntegration with metabolic states
| Fed state | Fasted state |
|---|---|
| High insulin | Low insulin |
| Active glycolysis | Active β-oxidation |
| Active PPP | Active gluconeogenesis |
| Active fatty acid synthesis | Suppressed synthesis |
03See the connections
The lesson names these links itself. Read the map left to right.
Inputs
- Acetyl-CoA (exported as citrate)
- NADPH from PPP
Machine
- Fatty acid synthase, cytosol
- +2 carbons per cycle
Controlled by
- Insulin, glucose, citrate (on)
- Glucagon, epinephrine (off)
- Liver and adipose only
Next: metabolic state integration.
04Reason with it
Four questions. Answer in your head first, then choose. You’ll see the reasoning either way.
Think first.
Why can’t acetyl-CoA simply diffuse out of the mitochondrion for fat synthesis?
Think first.
Which reducing agent does fatty acid synthesis consume?
Think first.
Is fatty acid synthesis just β-oxidation run backward?
Think first.
A drug blocks citrate export from mitochondria. What happens to fatty acid synthesis?
05MCAT lens
Predict the direction
High insulin → increased fat synthesis
High NADPH demand → increased PPP flux
Blocking citrate export → reduced synthesis
Fasting → synthesis suppressed
No memorization required.
06Common traps
Trap 1: Fatty acid synthesis produces ATP
Misconception
It consumes energy.
Correct model
Storage pathway.
Trap 2: Same enzymes as β-oxidation
Misconception
Completely different.
Correct model
Separate machinery.
Trap 3: Occurs in all tissues
Misconception
Tissue-specific.
Correct model
Division of labor.
07Dependency check
Can you reconstruct this concept without looking at the page? You should now be able to reason through:
Why fat synthesis requires NADPH
Check against 02 · C: NADPH is the reducing currency of synthesis.
Why synthesis and oxidation are separated
Check against 02 · A and E: different places and machinery prevent futile cycles.
How hormones enforce metabolic state
Check against 02 · F and H: insulin on, glucagon off.
Why PPP feeds lipid synthesis
Check against 02 · C: PPP is the main NADPH source.
If something feels unclear, the missing idea is usually thisFatty acid synthesis stores excess carbon and reducing power during abundance.
08Remember
REMEMBER · if you keep only a few things
- Synthesis is not reversed β-oxidation.
- Cytosol vs mitochondria; NADPH in vs NADH out.
- Acetyl-CoA leaves the mitochondrion as citrate.
- High citrate = abundance.
- On: insulin, glucose, citrate. Off: glucagon, epinephrine.
- Made in liver and adipose, not muscle.
09Forward hook
You’ve now seen glycolysis, PPP, β-oxidation, and fatty acid synthesis as separate pathways.
How does the body decide what to burn, store, or spare?
That leads to metabolic state integration.
